چکیده

Fungal Lipids, Ergosterol, Inhibition, Etaconazole, Fungicide The triazole fungicide etaconazole (C G A 64 251) interferes with the ergosterol biosynthesis in Ustilago maydis by inhibiting the C-14 demethylation o f the sterol nucleus. During the late log growth phase o f U. maydis a novel endogenous sterol metabolite (14x-methyl-ergosta-8,24(28)dien-3/?,6a-diol) was discovered and analyzed, which accumulates under the influence o f the fungicide. The structure o f this metabolite points to a hydroxylation-dehydration mechanism for the introduction o f the double bond at C-5 during the ergosterol biosynthesis.

Effects of miconazole and dodecylimidazole on sterol biosynthesis in Ustilago maydis.

Miconazole at minimal fungitoxic concentrations inhibits ergosterol biosynthesis in sporidia of Ustilago maydis by interference with sterol C14 demethylation. The action is analogous to that of the fungicides triarimol and fenarimol. The fungicide 1-dodecylimidazole at low concentrations (0.1 to 0.25 mug/ml) inhibits sterol C14 demethylation; however, at higher concentrations (1.0 mug/ml or gre...

A secreted Ustilago maydis effector promotes virulence by targeting anthocyanin biosynthesis in maize

The biotrophic fungus Ustilago maydis causes smut disease in maize with characteristic tumor formation and anthocyanin induction. Here, we show that anthocyanin biosynthesis is induced by the virulence promoting secreted effector protein Tin2. Tin2 protein functions inside plant cells where it interacts with maize protein kinase ZmTTK1. Tin2 masks a ubiquitin-proteasome degradation motif in ZmT...

Identification of a gene cluster for biosynthesis of mannosylerythritol lipids in the basidiomycetous fungus Ustilago maydis.

Many microorganisms produce surface-active substances that enhance the availability of water-insoluble substrates. Although many of these biosurfactants have interesting potential applications, very little is known about their biosynthesis. The basidiomycetous fungus Ustilago maydis secretes large amounts of mannosylerythritol lipids (MELs) under conditions of nitrogen starvation. We recently d...

Cloning and disruption of the ornithine decarboxylase gene of Ustilago maydis: evidence for a role of polyamines in its dimorphic transition.

The gene encoding ornithine decarboxylase (ODC) from Ustilago maydis was cloned. A conserved PCR product amplified from U. maydis DNA was synthesized and used to screen a genomic library of the fungus. Alignment of its deduced protein sequence with those of other cloned ODCs showed a high degree of homology. Gene replacement was obtained by removal of a central part of the gene and insertion of...

Characterization of the Ustilago maydis sid2 gene, encoding a multidomain peptide synthetase in the ferrichrome biosynthetic gene cluster.

Ustilago maydis, the causal agent of corn smut disease, acquires and transports ferric ion by producing the extracellular, cyclic peptide, hydroxamate siderophores ferrichrome and ferrichrome A. Ferrichrome biosynthesis likely proceeds by hydroxylation and acetylation of L-ornithine, and later steps likely involve covalently bound thioester intermediates on a multimodular, nonribosomal peptide ...
